Automated fluorescent dyer-based DNA sequencing provides data with very high throughput and is less hazardous than traditional radioactivity-based manual DNA sequencing methods. However, high cost of the instrument coupled with equally high level of expertise for its proper operation prevents procurement and maintenance of such instruments by a large number of organizations.

At GeneOmbio Technologies, we assist the scientific community by providing different DNA sequencing and ancillary supports in a rapid and cost effective manner using our Applied Biosystems- make range of automated DNA sequencing systems.

The different services include:

    Single strand, direct sequencing services
  • Single strand sequencing of PCR products up t0 550-750 bases.
  • Single strand sequencing of purified plasmid DNA up to 550-650 bases.
  • Single strand sequencing of plasmid in the form of Escherichia coli strain as a stab culture harboring the plasmid.

    DNA sequencing of DNA fragments cloned as an insert in a cloning vehicle
  • Single strand sequencing by ‘primer walking’ of the insert up to 1000 bp.
  • Double strand sequencing by ‘primer walking’ of the insert up to 1000 bp.
  • Single strand sequencing by ‘primer walking’ of the insert up to 2000 bp.
  • Double strand sequencing by ‘primer walking’ of the insert up to 2000 bp
  • Single strand sequencing by ‘primer walking’ of the insert up to 3000 bp.
  • Double strand sequencing by ‘primer walking’ of the insert up to 3000 bp.

Data generated through such studies form the base line for a wide range of down stream applications ranging from in-depth restriction mapping of the insert to detailed annotation of the DNA segment.

The data is provided as electropherogram with quality bars denoting the quality of base calls along with its electronic version for further analysis. We also provide BLAST analysis of the sequence and list of top ten hits on the public domain database with a copy of the sequence submission details of the 1st hit sequence. Translated hypothetical protein (using standard table) in all six reading frames along with restriction map of the sequence and list of non cutters is also provided in tabulated form. We also provide, free of cost, de novo primer designing of any segment of the sequence as identified by the customer.

Cloning and DNA sequencing services
With an objective to providing total solution to DNA sequencing services, we provide customized molecular cloning service prior to DNA sequencing. This helps in archiving of precious insert in E. coli and becomes an unlimited source for the fragment for further study. Since PCR is the most popular means of studying gene segments, we provide a rapid cloning service of PCR products prior to sequencing services.

Cloning and expression services
We undertake molecular cloning of coding sequences (total or partial) in expression vectors and express them as fusion proteins. The service includes cloning of the insert in-frame with the fusion gene, its sequencing to confirm the cloning junctions, optimization of the protein expression, its confirmation through western blotting technique using labeled antibodies specific to the tag-protein and providing the expressed protein in fused/released form generated at laboratory scale along with the recombinant construct and associated data generated from it.
